Introduction and Objective: We assessed the effect of CagriSema on eating control and behavior in adults with overweight/obesity.Methods: In REDEFINE 1 (NCT05567796), 3417 adults (67.6% female; mean: age, 47.0 years; body weight, 106.9 kg; BMI, 37.9 kg/m2) were randomized 21:3:3:7 to once-weekly CagriSema 2.4 mg/2.4 mg (n=2108), semaglutide 2.4 mg (n=302), cagrilintide 2.4 mg (n=302), or placebo (pbo) (n=705), plus lifestyle intervention, for 68 weeks. Prespecified secondary and exploratory analyses were changes in Control of Eating Questionnaire (CoEQ; 4 domains: Craving Control, Positive Mood, Craving for Savory, Craving for Sweet), and Three-Factor Eating Questionnaire revised 18-item version 2 (TFEQ-R18v2; 3 scales: Cognitive Restraint, Uncontrolled Eating, Emotional Eating) scores, respectively.Results: CagriSema significantly improved all 4 CoEQ domains vs pbo (Fig A). There were significant improvements with CagriSema vs cagrilintide and semaglutide for Craving Control and Craving for Savory, and vs cagrilintide for Positive Mood. CagriSema significantly improved TFEQ-R18v2 scores vs pbo and cagrilintide for Uncontrolled Eating and Emotional Eating (Fig B). Cognitive Restraint improved significantly with CagriSema vs pbo. No differences were observed for any TFEQ-R18v2 scales with CagriSema vs semaglutide.Conclusion: CagriSema significantly improved eating control, demonstrating both distinct and overlapping effects relative to its individual components.
